This study aimed to evaluate the harmful effects of metal accumulation on Astyanax lacustris and their biochemical response when exposed to the metals contained in the Doce River water, Brazil. The study was carried out in the Baixo Guandu region of the Doce River, Espírito Santo state. Samples were collected during the dry and wet seasons. Biochemical alterations were observed in the liver of A. lacustris, with apparent seasonal effects directly related to the high concentration of Al and Fe in this tissue. Despite this, the bioconcentration of metals in the organisms showed seasonal effects only in the gills, as they are the first organ in contact with the contaminated water. The data generated in this study provide an overview of the health of the ecosystem in this region, highlighting the harmful biochemical and metal concentration effects on the population of A. lacustris affected by the Fundão dam failure.

Ocean warming and acidification can cause deleterious effects on marine biota, which may be potentialized when associated with metal pollution. Thus, the aim of this work was to evaluate the effects of pH decrease, temperature increase and lead contamination on fertility rate and embryo-larval development of Echinometra lucunter. Gametes and embryos were exposed at pH 8.2 (control) and 7.5; at 26°C (control) and 28°C; and at lead concentrations of 0 (control), 125, 250 and 500 µg/L. These conditions were tested individually and in combination. The fertilization rate of E. lucunter was only significantly reduced in the treatments where temperature was increased and in the treatment where pH decreased. However, the development rate of the pluteus larvae was significantly affected in the majority of treatments: metal contamination in the higher concentration; decreased pH in all metal concentrations; increased temperature in the highest metal concentration; decreased pH and increased temperature and all variables combined, which is decreased pH, increased temperature and metal contamination in relation to the control group (C). The development test was shown to be more sensitive than the fertilization test in all the studied scenarios. In general, the present study suggests that pH decrease, temperature increase and metal pollution may have a significant impact on E. lucunter reproductive cycle.

The Lippia alba essential oil (EO) is a fish anesthetic immiscible in water and commonly used diluted in ethanol. We evaluated the effectiveness of surfactant use with Lippia alba EO in the anesthesia of Oreochromis niloticus, as well as its toxicity in fish and mammals. The EO was extracted by hydrodistillation and the fish were exposed to anesthesia at the concentration of 250 µL/L for 10 min with the surfactants polysorbate 20 (T20), polysorbate 80 (T80), polyethylene glycol (PEG), and ethanol. We also evaluated fish recovery and anesthetic safety margin after exposure for 10, 20, and 30 min. To assess the surfactants' toxicity in mammals, Mus musculus (mice) received the same treatments by gavage. The main constituents of the Lippia alba EO were linalool (42.36%), geraniol (12.46%), neral (10.7%), and limonene (7.45%). Deeper anesthesia was faster in the T20 (60 ± 2.9 s) and T80 (272 ± 21 s) treatment groups, while recovery time for T80 was longer (596 ± 47 s). All treatments showed a good safety margin, without mortality. The genotoxic effects caused by surfactants in mammals and fish were at similar levels to those found in the ethanol treatment. Therefore, this study demonstrated that the use of surfactants T20 and T80 in Oreochromis niloticus anesthesia presented neither a reduction nor a considerable increase of the toxicity when compared to the commonly used ethanol; however, an increase in anesthetic effectiveness was observed throughout the experiment.

In 2015, after the Fundão dam failure, in Minas Gerais State, Brazil, around 50 million cubic meters of sludge from iron mining tailings were discharged into the Doce River. After the dam collapse, surpassing concentrations of metals were observed in the river sediment, which could be harmful to aquatic organisms, including the fishes. The present study aimed to evaluate the toxic effects of both elutriate and water, collected from the Doce River, on Astyanax lacustris three years after the dam failure. A bioassay was carried out through subchronic exposure to Doce River water (E0) and three elutriate concentrations (10, 50 and 100%). Biochemical analyses (CAT, GST, AChE), metal bioaccumulation assays and calculation of the integrated biomarker response index, version 2 (IBRv2) were performed. The outcomes uncovered deleterious consequences on organisms exposed to E0, with AChE inhibition and bioaccumulation of Fe and Mn in both liver and gills. IBRv2 values were more elevated in fishes exposed to E0 for all tissues. Thus, the elutriate was not harmful for the assessed fishes, since complexing agents presented in the sediment, such as goethite and hematite, may have triggered metals' chelation. In this scenario, the elutriate may have acted as a protective agent for the subjected organisms, unlike the Doce River waters, in which contaminants were proven to be hazardous for the aquatic biota.

Manganese and iron were found at high concentrations (3.61 mg/L and 19.8 mg/L, respectively) in the water of the Rio Doce after the dams of Fundão and Santarém broke in Mariana/MG (Brazil). These same metals were found in fish and crustacean muscle (15 mg/kg and 8 mg/kg wet weight, respectively) in the specimens collected near the Rio Doce's outfall. Due to the variation in Mn concentration found in the lower Rio Doce, this study aimed to determine the effects of Mn in Oreochromis niloticus, at the concentrations allowed by CONAMA, and in concentrations found in the Rio Doce after the dams broke. The animals were exposed to the following dissolved concentrations: control group (0.0 mg/L), 0.2; 1.5 and 2.9 mg/L manganese for 96 h. In addition, a positive control was conducted, injecting intraperitoneally with cyclophosphamide (at 25 mg/kg). These exposures caused significant erythrocyte micronucleus formation in the organisms exposed to the highest concentration, as well a significant increase in the DNA damage index of erythrocytes from organisms exposed to 1.5 mg/L and 2.9 mg/L treatments. The glutathione S-transferase enzyme activity also showed a significant increase in the liver of the organisms exposed to 2.9 mg/L. However, catalase activity increased significantly in the gills of the animals exposed to all concentrations of manganese that were tested. Manganese bioconcentrated in greater quantities in the liver than the gills. Thus, manganese causes significant damage to genetic material, generates nuclear abnormalities, activates the body's detoxification system and can accumulate in animal tissue.

Considering the high number of accidents with diesel oil spills occurring in the marine ecosystem, toxicity tests aimed at assessing the effects of this pollutant on biota are necessary and urgent. Thus, the present study aimed to evaluate the toxicity of the soluble fraction of diesel oil (WSD) in the fertilization success of gametes and pluteu larvae of the sea urchin Echinometra lucunter. To do this, gametes and embryos were exposed to concentrations of 0% (control group), 0.5%, 1.5% and 2.5% of WSD. The fertilization success of exposed gametes and embryos were reduced significantly when compared to the control group in all tested concentrations. With this finding, it is evident that diesel oil can be significantly promoted in the early and adult life stages of a particular organism, and a better way of evaluating this toxicity is through the analysis of contaminant effects throughout the reproductive cycle of a species.

Mangroves are classified as permanent preservation areas and regarded as natural nurseries. However, they have suffered several anthropogenic stresses, resulting in their decline. In the light of that, comes the importance of researching their environmental characteristics and revealing possible factors that have led to the degradation of this important ecosystem. The aim of this study was to evaluate the environmental quality of different areas in the mangroves of Ilha do Lameirão Ecological Station through microbiological analyzes of sediment and interstitial water along ten (10) sites, distributed in two areas with different conservation levels (Canal dos Escravos (CE) and Maria Ortiz (MO)) between 2010 and 2012. The microbiological analyzes revealed that MO region, in all seasons of the year, achieved total coliform and thermo -tolerant coliform values above those permitted by the CONAMA Resolution 357/05, fitting the Class 2 conservation standard. The presence of high levels of total and thermo-tolerant coliforms in MO is a strong indicator of impacts originated from the human population and, consequently, the decline of the mangrove itself and the health of human communities surrounding that area.


Os manguezais são classificados como áreas de preservação permanente e considerados como berçários naturais, no entanto sofrem várias tensões antropogênicas, resultando em seu declínio. À luz disso, evidencia-se a importância de pesquisar suas características ambientais e reveladoras de possíveis fatores que levaram à degradação desse importante ecossistema. O objetivo deste estudo é avaliar a qualidade ambiental das diferentes áreas em manguezais da Estação Ecológica Ilha do Lameirão por meio de análises microbiológicas de sedimentos e da água intersticial ao longo de dez locais, distribuídos em duas áreas com diferentes níveis de conservação (Canal dos Escravos (CE) e Maria Ortiz (MO)) entre 2010 e 2012. As análises microbiológicas revelaram que a região MO, em todas as estações do ano, obteve valores de coliformes termo-tolerantes e totais acima do permitido pela resolução CONAMA 357/05, Classe 2. A presença de níveis elevados de coliformes totais e termo-tolerantes em MO é um forte indicador dos impactos originados da população humana e, consequentemente, do declínio do próprio mangue e da saúde das comunidades humanas que cercam a área.
